Set up the room
Comfort4Stimulation50+Higher Stimulation now lowers the chance the kitten inherits a parent birth defect, so keep it up on a support cat that has to live to act. Keep a dedicated breeding slot rather than crowding cats into a comfort-3 room. Run the line away from inbreeding, since inbred kittens get less out of Stimulation and inherit defects more often.

Cleric stays the single most important collar in the game through 1.1. Healing, revival, and Holy coverage are three separate ways to keep a run alive, and none of them got touched in a bad direction. Cleanse went single-target with a much larger upgrade area, and Open Wounds is now magic damage, so it scales off the same INT you are already breeding for. A Cleric/Fighter or Cleric/Tank core remains the safest opener at every difficulty. Run this collar first, every time.
