TL;DR
Wake Hypnos by crafting three Incantations at the Incantation Cauldron in The Crossroads, in order: End to Deepest Slumber, then End to Dearest Slumber, then End to Dumbest Slumber. Each one gives you +1 Dream Vapors. Gift one Dream Vapors to the sleeping Hypnos after each craft to drop Melinoe a layer deeper into his dreams. In the third and deepest layer, pick the Wake Up option. That rouses him and clears the Fated quest.
The catch worth planning around: the final Incantation needs Soot, and Soot only becomes available after you beat the game and watch the end credits. So treat this as a post-credits project, not something you finish on your first clear.

The short version of how it works
Hypnos shows up in Hades II slumped asleep in a corner of The Crossroads. He does not wake on his own, and no amount of poking changes that. Waking him is a Fated quest, and the whole thing runs on a single resource called Dream Vapors.
You make Dream Vapors by crafting Incantations at the Incantation Cauldron. Three Incantations earn three Dream Vapors, one per dream layer. You hand Hypnos one Dream Vapors at a time, and each gift pulls Melinoe a level deeper into his sleeping mind. Reach the bottom, tell him to wake up, done.
The order is fixed. You cannot skip ahead. Each Incantation unlocks the next, and the last one is locked behind a material you can only get after the credits roll. If you are still working toward your first clear, our Hades II beginner guide covers the run before you reach this point.
The three Incantations
Craft these at the Incantation Cauldron in The Crossroads. Build them in this order, top to bottom.

Every one of these grants +1 Dream Vapors when cast. So a full run of the chain leaves you with the three Dream Vapors you need for the three dream layers.
A naming heads-up, since the in-game spellings trip people up: the material is Shaderot, one t, not "Shaderoot." And the second Incantation is Dearest, the third is Dumbest. Easy to mix up at a glance.
Where to gather every ingredient
Most of these you will already have stockpiled by the time you are working on this quest. The two that gate progress are Mandrake and Soot.
Moly is the basic one. Gather it by hand out in Erebus, and it also turns up as an encounter reward during runs.
Shaderot grows in Tartarus and you can pick it by hand with no Gathering Tool needed. If you would rather farm it at home, you can grow Shaderot in the Crossroads garden.
Poppy comes straight from Melinoe's garden at The Crossroads. Plant Poppy Seeds, wait, harvest. No tool required, and it is cheap to keep a steady supply.
Mandrake is the first real wall. It lives in the Rift of Thessaly, and you need the Silver Spade to dig it up. If you have not unlocked the Rift or the Spade yet, that is your prerequisite before the second Incantation is possible.
Soot is the hard gate on the entire quest. You get it by speaking with Hades, alongside Persephone, in Tartarus, and that conversation only happens after you have beaten the game once and watched the end credits. Soot appears once that dialogue wraps. No credits, no Soot, no third Incantation. This is why we call the whole thing a post-credits quest. Walking through the dream layers
Once you have a Dream Vapors in hand, go to the sleeping Hypnos at The Crossroads and gift it to him. Each gift sends Melinoe into a deeper layer of his dream. There are three.
Dream Vapors #1 drops you into the first layer, a flat cardboard cutout version of the House of Hades. It looks like the old place rebuilt out of construction paper. Poke around, then come back to craft the next Incantation.
Dream Vapors #2 takes you one level down into a softer dream, a kind of pillow fortress. Cozier than the first, and a sign you are close.
Dream Vapors #3 lands you in the third and deepest layer. Here Melinoe finds a note, and the note more or less tells her to try saying wake up. Take the hint. Choose the Wake Up interaction and Hypnos jolts out of sleep with a yawn loud enough to wake the dead, which, given the address, is saying something.
What you get for waking him
Finishing the quest awards the Soundest of Sleepers achievement and completes the matching Fated Prophecy entry. After that, Hypnos is no longer parked asleep in his corner of The Crossroads.
